Zielsetzung

The interplay between excitatory and inhibitory neuronal networks is exquisitely balanced to maintain proper function of the brain. Consequently, severe imbalances may manifest themselves in pathologies such as epilepsy. At this point, it is unclear if certain neurons such as inhibitory parvalbumin cells play a more prominent role in maintaining the network balance. Such cell types would then be key targets for treatment and thus their identification is important in the understanding of some neurological diseases. We have developed an experimental pipeline that allows us to acutely increase or decrease the activity of specific neuronal cell types in the living brain. This is accomplished with various transgenic mice and viral vectors. Simultaneously, we monitor the activity of the network using fluorescence changes and in vivo two-photon microscopy. The goal of this thesis project is to manipulate different cell types and to analyze the effects on global vs. local network activity.
